Phase 1 trial reports high response rates with anito-cel CAR T therapy
A phase 1 study evaluated anitocabtagene autoleucel (anito-cel), a BCMA-directed CAR T-cell therapy using a synthetic d-domain binder, in 38 patients with heavily pretreated relapsed or refractory multiple myeloma. The trial reported that all patients responded, with 79% achieving complete response, median progression-free survival of 30.2 months, and 36-month overall survival of 65%; grade 3 or higher cytokine release syndrome and neurotoxicity were rare. Complementary in vitro studies found the ddBCMA binder had a faster off-rate than a comparator dual-VHH binder and was associated with less cytokine release without loss of cytotoxicity.
